The city Dany captures at the end of S3 has 200,000 slaves, so that would easily boost the total population to above that of King's Landing. Which isn't that surprising. If we compare Westeros to Europe in the Middle Ages, Essos would be during the time of city-states and land empires. Rome had a population of around a million at its height.
The Dothraki crushed the Qohorik heavy horse, and the sellswords in the city's service fled. Three thousand Unsullied, slave soldiers from Astapor, withstood the Dothraki charges, however, and saved the city. Since the victory of the famed Three Thousand of Qohor, the city's defenses have been mostly left to the Unsullied.
